All,
I accidentally connected a battery in reverse polarity to my '06 250 ie GTS.
Now, there's power being delivered to any system in the scoot'.
Fuses in glove box are okay and pass resistance test. Can see the elements in the fuses are okay as well.
Any ideas?

The main 30A fuse will have blown. By the seat hinge, under the pet-carrier. Pages 54 to 57 of the user manual.

It good to see, when someone write again when the problem solved. I don't like when someone write only the problem, and later doesn't write the result. I would like to learn, and I want to know if me or someone else helped.

I agree. This was a simple one. One problem, one solution offered. Problem solved and acknowledged. Sometimes many solutions are offered, the problem gets solved, but I can be left wondering which solution(s) worked.

Can I ask why the 30Amp fuse blows when doing this?
(just curious)

With the battery polarity reversed, a huge current will back-flow through the regulator and blow the main fuse before any further damage is caused.

A new regulator/rectifier will arrive tomorrow and will be installed, hopefully, in time to ride the scoot' to a race at Sonoma Raceway on Saturday morning.

Was the regulator damaged? Usually it isn't. I've had to deal with several instances of this on several different Piaggio models, and in no case was anything damaged apart from the main fuse.

I don't know the root cause but the voltage across the battery when the engine is running is between ~15.5 - 18.6 volts. This may have contributed to the damage of another battery that was in the scoot'. I'm reading that the regulator can fail and that the above voltage is evidence that this one is damaged and needs to be replaced.

Yes, that voltage means the regulator is toast, and needs to be replaced NOW.

This is probably why you found you had to replace the battery in the first place - a duff regulator fries a battery in very short order.
