I'm taking a fresh look at the mixture on a scooter and I've put in a new spark plug to do a color check...about how much run time should I allow before I can use its color as a proper gauge? I know what to look for but I was wondering how long others wait (if at all) beyond a single ride.

Easier to get a plug to change color then get color in my experience.

Get can take hours.
Change is almost instant.

If you have an existing plug, consider using.
Shoot pic of before.
Use daylight and no flash.
Doesn't have to be direct sun tho.

Then shoot "after" pics.

charlieman's not wrong. Seems to take about an hour of runtime to get a useful reading on a new plug. The color takes a while to build up.

Better to use a recently used (but not worn) plug. Clean it with a soft brass wire brush and give it a good wash with brake cleaner and compressed air. It'll pick up color right away, after just a few runs.

That said, you need to use a fresh plug if you're planning to do a plug chop.

Good rule of thumb is about 30 miles in one outin then check every 20+ miles after use your older plug and don't keep cleaning it after a reading you are trying to build up a colour after all !! New plugs will take a lot longer to build up a true reading colour !! You know what your aming for colour wise !!
