Basic coolant service...? Lower hose clamp only. Here's why. 1. the three phillips screws can be a bear to get off. 2. you only miss a shot glass of liquid by leaving the cover in place. 3. no chance of having a gasket problem if you don't remove it 4. less likely to disrupt the other two hose clamps causing leaks. The only time I pull that cover off is when removing the whole water pump housing.
